DNS is the domain name service. It is a bunch of machines spread around the internet that hold the mapping between domain names and ip addresses. If you request a page, your browser will query the nameserver for your ISP, and get back an IP address which they can then contact directly to request the page. If your page won't load due to DNS problems it could either be your hosting provider hasn't set things up properly, or that peoples ISPs havn't set things up properly.
Also, be aware that a newly registered or newly transferred domain name will tkae several days to propagate through the DNS system. (They periodically query each other to update their info)
