I have been using two programs that I like for publishing our newsletter. One is GroupMail and the other is PostMaster Suite. You can use the online services that they offer or use the PC version. I really like PostMaster Suite. It will manage all the subscribes and unsubscribes and can be used for deleting bounces or invalid emails addresses. It uses Microsoft Access for the database. I just fried my Access, so I'm doing it manually until I get it up and running again.
When I find a program that I like, I usually become an affiliate or rep for them so I can sell it from my newsletter or site. Might as well recoup some of the money spent.
I'm also a member of a couple of ezine co-ops. If you want to ad some subscribers to your newsletter, there are several that are worth checking out. The bad thing about them is that you definitely have to add new subscribers manually since that new subscriber will get one free ad in the bargain.
